Custom DNA Cloning for expression of an N-terminal epitope-tagged or fusion protein in mammalian cell lines.

Epitopes include FLAG, MYC, HA, and V5 for immunological detection and eGFP, eYFP, DsRed, and mCherry for fluorescent detection.

Use the Design as You Order system to choose a promoter (CMV or EF-1 alpha), selection (Neomycin, Blasticidin, or Hygromycin) and tag. Simply enter the name of your construct, the DNA sequence of your coding sequence (or upload a fasta file with multiple sequences), select your choice of promotor, selectable marker, and C-terminal tag.

Important: The DNA cannot have any spaces, must be a multiple of 3, and be less than 5000 nucleotides (see Description below).

DNA cloning made simple with Design as You Order

Our comprehensive DNA cloning service allows users to simply enter the DNA sequence of the coding sequence (CDS) insert, choose a N-terminal epitope tag (FLAG, MYC, HA or V5 or fluorescent fusion protein (eGFP, eYFP, DsRed or mCherry), a promoter (CMV or EF-1 alpha), and the desired mammalian selection (Neomycin, Hygromycin, or Blasticidin) from the dropdown selectors. The  insert CDS will be cloned into a vector with the functional elements shown in the image.

Notes on Ordering: The DNA of the CDS must be a multiple of 3 and cannot have spaces to ensure the proper reading frame is designed. Adding the start (ATG) or stop (TAA, TGA, TAG) codons is optional.  Multiple DNA sequences contained in a single fasta file will contain the same vector elements chosen for that order.  If you have multiple sequences and want different elements, these should be entered separately.

Based on your choices, a vector will be designed, and a pdf will be sent via email prior to the start of synthesis.  If you notice any issues with your vector, please contact customer service within 24 hours.  Apheon Bioscience is not responsible for errors input or uploaded by the user after production begins.

Currently, clones are delivered as a bacterial stab along with a plasmid DNA at 100 ng/ul.
